Excel file vba take long time to run

Hi all,
looking for some expert advice as I have searched everywhere but no success.

I have a large excel file ~ 80b. The file has lot of formulas and not optimized to begin with.

So while I understand the opportunity to fix the file, the THING that is bothering me is that the file takes any between 2 to 8 hrs depending upon machine to machine and day to day. This is the SAME file, all the machine have similar configuration.

I am trying understand as what system resources or excel settings impacting us that the run time is different most of the time (some times on the same machine too)

Appreciate if anyone can give any guidance so we can attain same run times.

Excel Facts

Copy formula down without changing references
If you have =SUM(F2:F49) in F50; type Alt+' in F51 to copy =SUM(F2:F49) to F51, leaving the formula in edit mode. Change SUM to COUNT.

Also, I mentioned - this has something to do with configurations /settings of Windows and Excel. I already know that my files needs to be fixed.
