What does your data look like?
Are you looking for a perfect match of "MyText" for each cell, or do you want to count it if "MyText" appears anywhere in the cell, i.e. ("Something something MyText something")?
If you are looking only at exact matches, then you should just be able to use the COUNTIF function to do this (https://exceljet.net/excel-functions/excel-countif-function).

In range above there is text (Mytext) in some cells not with combination like (????Mytext???)•

It looks like you can still use COUNTIF, i.e.

=COUNTIF(E4:E500,"*MyText*")

Note, if you want to use this function in VBA, you can do it like this:

Application.WorksheetFunction.CountIf(...)
